I have an LCMS/MS assay to detect human parathyroid hormone (PTH 1-34) in the plasma going. My LOD for PTH(1-34) is as low as 500 pg/mL.

But when I conjugate the human parathyroid hormone with Alendronate (a bisphosphonate compound) at 13 amino acid (see the picture attached) I loose my sensitivity so much so that I dont see any peaks at concentrations  lower that 10 ug/mL.(see attached PPT file)

Firstly, what Alendronate have done so wrong with the fragmentation and ionization process to mess up the who LCMS/MS assay and its sensitivity to drop from pg to ug.

Secondly, what I can do to work my way around this problem and actually qualify the Alendronate -PTH-conjugate in the plasma samples (any other method)?
