Hi Kasturee,

one thing when using a microplate instead of a cuvette is that the volulme has to be equivalent in all wells. In a cuvette, the transversal light pathway if always of 1 cm, while the vertical pathway can vary when using a microplate if the wells are not filled with the same volume. Attached a document on pathway correction.
