As you know, cells are placed in incubator that has 37 oC with 5% CO2. If you do not pre-heat your media to 37oC (same temperature with the incubator), you will create stress to cells and possibly affect their 'health'. So it is important to prevent creating further stress since they have already contacted with trypsin/edta and detached from the surface of the tissue culture plastic which creates stress to the cells. I hope this helps.
Sedef thank you. I can now understand the reason why it is important to pre-heat the media. In my understanding I thought of the temperature (37oC) in relation to the body temperature for human body. It is not applicable??
In my case, I am working with cancer cell lines, whose once taken from humans, so your thought makes sense. However, I do not know i.e insect cells etc.
