After using an AChE enzymatic histochemistry (for slides) protocol (attached below), we observed some precipitates in the tissue when observed under the microscope. We checked the reactives used, the solutions used, but the result is the same. We also covered the beakers in which we made the solutions for the incubations and the enzymatic reactions with parafilm in order to avoid contact with the silver paper we use to cover them from light.
Does anyone know what else could be interfering into our histochemistry that can cause these awful precipitates (picture attached below).
So - if I am allowed to ask honestly - what was the solution of the problem shooting besides | Can you attach protocol? | being marked as the >best answer
After having troubles to upload my answer to Laia Vila from my RG-iPhone-App properly (had no feedback about my 'adding my answer' for 4 times...but was able to delete those 4 identical answers via iPhone...a 5th time-varied answer I was not able to post any more after that). I apologize if I missed the "Answer, regarding trouble shooting" since it is unusual to present an answer in an attached doc (BTW...in my iPhone-RG-App I can't see - or better - I cant read the Attachment....perhaps due to format .docx instead of being a pdf (but perhaps this is only a problem to me...old guy...not very familiar with using the RG-App on my iPhone). @ Laia Vila : I am a bit confused - having read your protocol, attached to this thread as "Protocol_AChE_adaptat a porta_040 520_ok.docx" (which seems to be ok ), I can't find there any hint on answering the ORIGINAL question, which was: (quote):

(end quote), which suggests that you were seeking for an answer (from) what the precipitates initiially came from / were induced and if there is someone out WHO is hable to comment on HOW such precipitates can be avoided.
Again: it is quite unusual to find i) Milorad Dragic 's question "CAN YOU ATTACH PROTOCOL" to be endorsed as the BEST ANSWER, ii) the Original QUESTION indicated to being SOLVED, without any further consideration or describing the improvement(s) or answers given, and, last but not least, iii) to end up with: "I have attached protocol" | "I've already attached it. I hope it can help you!
So - in the end - again my question (it is not for my personal satisfaction, but only for others looking into your initial question and honestly demand your 'solution' :
What was the culprit of your staining problem proper (= formation of such ugly precipitates...)? TIA WHM
