I currently conduct a toxicity test using zebrafish adults for my methanolic plant extract. I have 100% and 71%cumulative mortality for the fish at the concentrations of 250mg/L and 125mg/L, but my 1000mg/L, 500mg/L, and 62.5mg/L were all fine.
Is there anyone who has a similar experience? what could be the possible reason?
(All water quality parameters were checked and were normal)
This is likely due to an error. For example low number of fish per chamber increase heterogenity of the fish population among the test unit.

I used 7 fish in a tank. All the concentration had the same number of fish. Do you mean that might because the fish I used in some of the tanks not being healthy already?
I tried to repeat the problematic treatment group, but the same result was obtained. The mortality was still higher than the highest concentration.

Can you please tell more about mortality in control group (if any)? Are you getting the same reslults across all the replicates?
Possible reason could be due to wrong solution preparation- You can prepare a new set of solutions. If chemical is water insoluble then prepare the test solution in a suitable solvent. Forget not to keep a solvent control.
Yeah, I would start with remaking the solutions. Solvent-only control has to be included as well.
