Diffrent solvent use for same plant (from non-polar to polar) or pentane to water extracts, will never show same results because every solvents will extracts different compounds. e.g hexane will extracts fats or waxes, chloroform will extracts alkaloids, methanol :water combination will extracts phenolics etc. Due to difference in the polarity of solvents different compounds are separated. These different compounds have different scavenging capability. So in vitro free radical scavenging activities will be different. Higher in low polar solvents and maximum in polar solvents (phenolics due to -OH groups).

Hi Mr. Wang , Is that necessary to do TLC for my plant crude extract... Since i do not know solvent system for preparative TLC to get seperate spots on my plate...

You might have colloected one plant from different localities. this so happen that the same plant collected from different localities or different altitudes have different scavanging and antioxidant activities.
I agree with Robin Joshi, activity depends on the type of metabolites present on extract and also depends on the assay you carried out. Most of antioxidant assays detect polar antioxidants (flavonoids and phenolics are good candidates) but if you carry out tests as the conjugated diene assay you can also detect non polar antioxidants. Activity depends on metabolites content of the extract and kind of test carried out.
