Sometimes water vapor appears in agar plates even after the agar has been complete solidified, cooled to room temperature and dried in laminar air flow.I think the trapped moisture and vapor is due to the temperature shift from laminar flow in the incubator - am I right, or is there another reason?
Becouse the plats closed immediately after pouring the agar so let agar for few sec. Befor pour it or letthe cover of plat not closed very will
Some time water vapors are being produced by microbes as by products. As the cells multiply and colonies grow, water vapor will be produced . If the plates are in the normal position, than vapor will collect on the top lid. Once enough vapor has collected, drops of water will drop down onto the colonies, washing them away into one big mixed colony, leaving you with very little well isolated colonies. So, store them inverted so that water droplets can't ruin your well isolated single colonies!
I would also like to add upon other comments is that one can stake plate over other than only the bottom plates will have some condensed water.
Secondly, incubation conditions has to be monitored.
When does this occur? After pouring them? My experience here is, that this occurs, when the plates are closed to early. The hot air above the liquid agar can hold much more moisture, but when the plates cool down, this water condensates on the surfaces. In my experience its not necessary to pur and dry plates in the laminar flow when you have antibiotics in there. Doing this on the bench with not to much disturbances works fine.
Plates can get wet in the incubator when they are much colder than the inside of the incubator, since moisture from the incubator can condensate on the cool plates.
Neveen got it! do not overdry them and store them upside down in refrigerator.
you must pour agar under 40 degrees,after solidification of your media. keep them upside down in all your handling,
I think the reason is the shift of temperature within the room or incubator and also the immediate close after pouring the nutrient agar.
