I have performed a goldner trichrome stain to perform histomorphometric analysis of the metaphyseal zone of MMA embedded bone. But I fail to stain the mineralized metaphyseal bone, the rest of my bone sample isn't stained very clearly as well. I followed the correct protocol and looked at amendements, but can't find the problem. Can someone provide me with some help? Thanks in advance.

please allow me to comment on your Masson/Goldner staining.
Masson/Goldner is a multicolor staining that provides very beautiful and informative staining. Especially for bones it is a very helpfull techniques as so many cell types localize in bone close to each other. However, Masson/Goldner is not appropriated for quantitative questions due to variable colouring.
Variability comes from MMA embedding procedure, deplastification, fixation of tissue, sample origin (mouse, human), thickness of sections, staining chemicals, and last but not least the specific staining procedure.
I attach our Masson/Goldner staining procedure. Hopefully, this is a good starting point to improve your work. Please find papers with further examples.
If you require further information please contact me. For what purpose you wish to stain bones with Masson/Goldner? Might be there are other easier options to achieve these goals.
