Why does histone requires short expression induction time?

Hello all,

I'm planing on a histone related research and found multiple papers describing expression induction conditions. All of them induces histone expression at 37 degree C for 2~3 hrs. Is there any reason for inducing only for less than 3hrs? wouldn't expression yield be better if the induction period is extended?

Also, some articles mention [pET-Histone expression plasmid] but I couldn't find a manufacturer of such plasmid. Does anyone have any information about such plasmids?

Hi, Apparently you are talking about recombinant protein purification of histones. If that is the case, then here are the answers;

1-Induction time/temperature vary for different proteins. In this case, 2-3 hrs induction gives you enough protein for purification.

2-pET vectors are base vectors in which you can clone your gene of interest for protein purification. Here is the link related to the vectors. you can buy and have a stock of your own. various versions are available, select the one that suits you the best.
