I think just because of the gel is easy to break down at the next step of proceed in the method. And you can not store the gel for long time, but you can do that with nylon membrane. I recommend that you should replace nylon membrane by polyvunylindene difluoride membrane

how long you keep your gel the bands will get diffused, as Trung said PVDF membrane is the best and most widely used membrane now a days.
