I am growing Anabaena sp. PCC7120. They seem to grow well on agar plates containing BG11 and BG11o. However, they die after 3 days when I transfer my cells to liquid BG11 or liquid BG11o. I am growing my cells at 28C under 50 umol/m2.s PPFR and gentle agitation of 80 rpm.
Plz, optimize the growth of the organism in respect of temperature, pH etc.
We are regularly growing Anabaena sp. PCC7120 in liquid BG11 without problem. I would suggest increasing the NaHCO3 concentration to 10mM, as CO2 starvation might be an issue in liquid more then on solid media (due to surface to volume ratio)

Thank you all for the suggestions. I realized there was metal precipitation after autoclaving. I also adjusted the pH before and after autoclaving the BG11 and it is working. I will try increasing the concentration of bicarbonate as Mr. Yoram suggested. I appreciate your help.
Initial growth need more inoculam. Add culture in low volume of media initially, with low light and without agitation. Once the culture stabilise and grow then, you can shift it to high light and what ever conditions you like, it will tolerate
