I am taking some measurements using a spectrometer connected to a microscope. My aim is to know the spectral position of the resonant peak. As I know, I need to calibrate the spectrometer for bright state and dark state. I don't know the reason for the calibration.
The quality of the calibration depends on the application. The bright and dark images you are taking sound like a type of flat field calibration which will correct for the spectral dependent transmission through the microscopen and the light source. If you are making quantitative measurements for intensities at different colors then you would need to know how much of an affect the microscope optics have. The spectrometer and the detector also have wavelength sensitivities. Correcting for all of these can be difficult if it must be very accurate and NIST traceable, but in most cases just taking a bright and dark image can be used to then correct the image in post processing.
