In the real-time experiment usually the melting peak is about 85 degree celcius. But I found for one of my gene of interest higher-Tm peak about 94 degree celcius.
Has anyone observed this phenomenon and can give an explanation?
Dear Seyed, it may related to amplicon size and GC content of your amplicon.
Dear Mert, you are right. Thanks
