I did recently glycogen test on liver where we found that glycogen conc. was significantly less than control diet rat. here , we used high carbohydrate high-fat diet rat model. We were expecting that glycogen conc should be higher than control diet, however, we got opposite result. any explanation will be highly appreciated.
If high fat and high carbohydrate diet was fed to animals long enough to accumulate substantial fat in their liver, cause hyperinsulinemia and insulin resistance, you should expect a reduction in glycogen contents in livers; it is the normal pattern.
I agree with dr. Tausif. You can check lipid accumulation with the fluorescent lipid proble, Nile Red (on frozen sections).

Many thanks for kind reply. I have only paraffin wax fixed tissue samples. I am looking for alternative staining method of Oil Red O to see lipid accumulation. Would you recommend any staining method for lipid deposition other than oil red O.
