I am working on phosphorus analysis in water using molybdenum blue method. I use the method explained in the paper “Test of an Ascorbic Acid Method for Determining Phosphorus in Water and NaHCO3 Extracts from Soil . F. S. WATANABE AND S. R. OLSEN, soil science society 1965”. I have several questions about it.

I do not get good repeatability in the absorption of standards i.e. I obtain different absorptions in different days for standards( the standards with color development are prepared as fresh ). I think the intensity of color is differenet in different days. Also, the color of blank, standards and samples becomes more intensive quickly after reading of the absorptions.

Two users said that I should acidify the standards and samples before adding the molybdate solution. But the standards( potassium dihydrogen phosphate solutions) are acidic i.e. they are colorless in the presence of phenolphthalein indicator. Nevertheless, I added several drops of 5 M sulfuric acid. However, the amount of absorptions was high and the colors of both standards and samples became quite strong blue after reading absorptions.

I would be grateful if you guided and answered me.

Thank you in advance.
