I ran a gel with protein from SH-SY5Y cells. I used BCA assay to determine protein concentration so apparently I loaded 30ug of protein. I used a stain free gel to visualize total protein content. There is only a faint band in the middle of my gel. I went ahead and transferred the gel and visualized the membrane and had the same results. When I do ponceau staining for the membrane, I get the same thing. No bands except for very faint bands in the middle of my gel. This is a precast gel so nothing wrong there. What is going on?
