Why am I not getting any positive colonies in my transformed GV3101 Plate?

I had transformed positive plasmid in GV3101 and got colonies after 2 days. But I did not get a single positive colony after colony PCR. What will be the reason? Although I used required antibiotics (rifampicin and kanamycin).
