I am doing sagittal mouse brain sections of thickness 300um from postnatal days P20-P24 and P47-P50.  Sections are sliced in choline-ACSF, incubated at 34C for 15mins before transferring to RT.  Leave them at RT for atleast 30mins prior to recordings.  I use a thin walled pipettes and a tungsten bipolar electrode for stimulation.  I placed the recording pipette in CA1 dendritic layer and the stimulating electrode in Schaffer collaterals, positioned 250um apart.  I have tried placing them further apart and brought them as close together as possible. Nevertheless, I do not get a synaptic response.  Attached is the response that I have seen so far.  The response I received does not get blocked by any receptor antagonists suggesting that it is not a synpatic response. Do any of you have any suggestions on what am I missing here and what all could have possibly be gone wrong?
