I used alcoholic extract of peganum harmala seeds as inhibitor to staphylococcus enterotoxin type A gene (SEA). By using diffusion method I need to know how can i take a smear from zone of inhibition to know the effect of extract on SEA gene ?
Is there a reference for evaluating substances against toxins by diffusion on agar?

I have taken smear from the zone of inhibition on an agar plate several time for culturing and microscopic observations.

For DNA extraction, you need a lot of cells. I do not think that you can have them in the zone of inhibition. However, you can try sub-culturing the smear from the zone and extract DNA. It depend on the mode of resistance developed in the bacteria to grow in the zone. It it is not reversible, then you would be able to have culture of the resistant clone only.
