I would like to store the fresh leaves for a time until use. How can I preserve it as I would like to separate the chloroplasts? Could anyone let me how the method(s) to do that?
You can store the leaves if you intend to isolated thylakoid membrane or intact chloroplast in around 14-16 oC for around 3-4 days under wet condition. I have isolated the intact chloroplast from the stored spinach leaves and it worked well. But always isolation from fresh leaves is preferred for enzymatic activity as there is high chances of protein degradation and thus loss of enzymatic activity under such condition.
Depends on what you want to do with the choloroplast once you have stored it - can you be a little specific? and also exact time that you are looking for.
For isolating intact chloroplasts from leaves and if requires storing before isolation the samples can be stored for less than 40 hours in refrigerator, within a Petriplate and under light.
