As i read many paper of biofilm all where mention that by taking optical density we can say that either week biofilm or strong biofilm but if we see the film on the microtiter plate how we can take the optical density? kindly tell me the stage where i can take optical density.

It stands to reason that as a biofilm develops, the biomass will increase to the point at which no further growth occurs (e.g., because of resource depletion) or because physical disturbance begins to take a toll on the structure.
If you want to have a specific 'time' to analyse your biofilms, it is probably good to first run a time course experiment four your strain(s), growth media, growth conditions and temperature, and then choose the best time that gives you useful results.
If you leave the measurement too late, many o the cells in the biofilm may no longer be viable, and some of the biomass might be lost to physical disturbance. Some bacteria also release surfactants to allow cells to escape the biofilm as part o the maturation process.
If you do the measurement to early, then there may not be enough material to give a significantly-different measurement compared to the sterile control.
You need to chose a time for your measurements that is just right - as in Goldilocks!
