While taking OD of a microbial suspension, if OD values are more than 1, is there any problem with reliability of data?

I am talking about the digital spectrophotometer (and not the analogue type). Theoretically OD = 2 - log T, thus max OD can be 2.

While quantifying microbial growth at 625 or 660 nm, if the OD reaches above 1, how much necessary is it to dilute it to bring OD between 0.3-0.7?

Hi, Vijay,

There won't be a problem with reliability of data. OD600 more than 1 is fine but to what extent beyond 1 is something to be considered. Ideal range of any reliable spectrophotometer whether Digital or Analog should be 0.2 to 2. But ensure that you don't exceed beyond 1.5-1.8.

If your values go beyond this then certainly you have to dilute the culture.
