I'm planning to use the diameter of an eye pupil in order to provide some parameters to adapt the Graphic User Interface according the diameter of the pupil.
I think you can measure how tired someone is by looking at their eye. There was a researcher team here in Germany who created a device to measure how fatigued divers were on the rest spots of the autobahn. I am not sure if they had to stimulate their eye and look at their iris's reaction speed or measure the length of the blinks or a combination, but you can try to find out about their research to get you started.
In my opinion, pupil diameter can tell us about location of object in focus. Large diameter indicates that object of attention is far away while small diameter indicates that object of attention is near eye.
