I am trying to macerate fibrous and voluminous plant tissues, such as stems and roots, and I am facing some problems with that. We have been using a bead beating grinder, but the low throughput when using 50 mLs tubes is a major problem, besides leakage and vial cracking. We macerate the samples with a extracting solvent, and I wonder if it would be possible to, instead of using the whole material (e.g., all of the root material of one plant), taking a smaller amount, and normalize the results by the weight of this selected material would be enough to accurately represent the whole original sample. Could anyone help me with ideas of how making the process of maceration of this kind of material easier and more efficient?
