Multistep extractions are more effective than a single step. it could be a better way to separate the pigment from the extract first. After adding specific chemicals into the residue, a mechanical treatment (such as sonication) is required and then centrifuge the sample.

The chemical materials, is better, to add after separation of the dye. Because the solvent contains a large number of extracted compounds which interfere with the added chemicals.
