If in colitis mice model most appropriate samples to check cytokines from would be colon tissue since it is the site of inflammation. Having said that serum can also be used to test cytokines at systemic levels. I have been using Bio-Rad Biolplex kit for cytokine analysis from Colon tissue as well as serum.
