I am looking for an RNA kit to extract RNA from cells. This RNA will have to be high quality and good yield for RNA seq analysis. Suggestions? Thank you
For cleaning up and concentrating RNA I would highly recommend ZYMO RNA Clean & Concentrator - made my life a lot easier for library preps. The best method for RNA isolation really depends on what your cells are, personally for our amoebal cells we use TriReagent followed by Zymo column. For the bacterial RNAs it is generally a version of Qiagen. A to yield these days you do not need huge amounts of RNA - even the Service providers have reduced their minimum requirements as the new kits come out. If I were to focus it would be on quality but if the source is limited there are ways around 'poor' quality that will still give decent RNAseq results.
Please provide more details of what type of cell you are working with . Like viral , bacterial , parasite or, cell culture etc. I am working with virus culture, I am getting good quantity and quality of RNA with viral Qiagen RNA extraction kit....!!!
We obtained very good results using "trizol (Gibco) and RNeasy columns (Qiagen)" for bacteria and "RiboPureTM-Yeast kit (Ambion)" for Saccharomyces cerevisiae followed by rRNA depletion with specific kits like RiboMinusTM Transcriptome Isolation Kit (Invitrogen).
PS: for more details about the procedure see "The transcriptional landscape of the deep-sea bacterium Photobacterium profundum in both a toxR mutant and its parental strain" (PMID: 23107454).
I have used QIAGEN's kit that lets you isolate, both RNA and protein from mammalian cells. This was specifically recommended by our Core facility, when I asked the same question. My experience was good with RIN scores between 9.8 - 10.0
