I want to perform immunohistochemical staining for rat chondrocytes but I do not know which primary antibody I should choose for detecting collagen type II. Can anyone offer recommendations?
I have used the CIICI antibody from the hybridoma bank and had very good results. Make sure you fix with alcohol and not PFA
II-II6B3 from DSHB. Works beautifully. I am attaching data sheet.
Thanx David and Luz for your suggestions. Actually I am looking for the primary antibody that is presented with TBS, in place of PBS. I searched for several days, but I couldn't manage to find it out. If you have any information about that, please let me know. Thanx again to both of you.
I've used Calbiochem CP18 (Mab) and works beautifully in paraffin sections, but had to use Chondroitinase and Hyluronidase to retrieval of epitope. Abcam (ab34712 - rabbit poly) also works, but just for intracellular staining.
Can you also share your protocol for antigen retrieval with chondroitinase and hyaluronidase? Thank you very much in advance.
