I would like to clone and express a segment of viral RNA in C6/36 mosquito cells but do not know which plasmid to use for the experiment. I tried using a plasmid that I use in mammalian cells with a CMV promoter but didn't see any GFP signal upon transfection into C6/36.

1- you should check the transfection efficacy with Mock GFP vector .
2- optimized RNA sequence in order to mosquito codon usage for increasing rate of recombinant protein expression .
