There are no reported physiological tests that can reliably distinguish these two species. Philippe Poindron is right that the presence of a parasporal crystal is the conventional defining mark of B. thuringiensis. Isolates of B. mycoides typically form very distinctive rhizoid colonies on agar. Keep in mind that other Bacilli can have these characteristics. Both B. thuringiensis and B. mycoides have 16S rRNA gene sequences that place them in the Bacillus cereus group of species, however.
