Root tips obtained from the plant grown in land or in a pot, root tips from the seed germinated in petri dish, tips of very young leaves, flower primordiums, growth points of the buds of lateral branches, very young petals, glumes of some monocotyle plants can be used for the examination of somatic chromosomes. Root tips are the most commonly used part among all these, when conducting a mitosis examination. However, in a study on birch (Betula L.), a reliable method for preparing metaphase chromosomes from plant leaf tissues is described. The chromosomes were suitable for karyotype analysis. Compared to root-tip chromosomes, leaf chromosomes tend to be longer, or less condensed, and hence more karyotypically differentiated. Metaphase index in young leaf tissues was also very high. Metaphase spread consisted of evenly and well-distributed chromosomes and this allow accurate counting.

In my experience root tips are the most ideal. Very often, the problem is how to raise good, intact root tips that are of the right size and quality. The right size is root tip that is not too big such that too much meristematic tissue is not involved. The right quality is tissue that is young, tender and not woody. You can raise root tips straight from seeds by germinating in Petridish or in loose medium like vermiculite, saw dust or charcoal from rice husk. All these media allow root harvest without damage. If your primary root is large try decapitating it and see whether multiple and more slender secondary roots are formed. You can of course induce rooting from slender branches by marcotting if the seeds fail to root efficiently. You can also try the leaf tips.
