For bacterial study which one organism was use full - isolation and identification of microorganisms from sewage sample, soil sample.or getting from a organization like ATCC, MTCC.
It's better to get the organisms from ATCC or MTCC because they have isolated and have the purified colony without any contamination. If you are trying to isolate and maintain it is a time taking process...
That was a smart work. However, On the contrary if you try to isolate and identify the strain in your own Microbiology lab, then you ll have a complete knowledge on the same and you can study everything about it.
Dear Rajkumar, your question has two possibilities. First, if you bye a microorganism from ATCC or MTCC you will have of course a characterized strain that you can use as a control reference in your experiments. On the other hand if you isolate a particular strain of m.o. it may or may not has a unique features and you can compared them with a type strain like ATTC and notice a difference. Good luck!

It is worthy to use both referenced microorganisms (ATCC, MTCC,...) alongside with the isolated microorganisms from your sewage samples. It will give a great value to your study.
It really all depends upon the studies you are intending to do. If you goal is to characterize a known pathway or process then you should use an established strain, if your goal is to identify microbes with unique metabolism to solve a problem then you might be best served by screening natural isolates.
