As per the tiltle said, I want to qualitatively test how much IAA can be produced by some of the bacteria that I have just isolated from a soil sample. After culturing the soil bacteria in a liquid medium, I will spin it down and mix its supernatant with Salkowski reagent and then read its absorbance at 530nm to determine its IAA concentration.

However, several literatures I came across suggested different culturing liquid medium such as YMB (Yeast Mannitol Broth), LB broth, Nutrient Broth, or TSB (Tryptic Soy Broth), so I'm unsure which one is the best.

Even the suggested concentration of the L-Tryptophan that will be added into the culturing medium also varies from 0.5 g/L to 1.0 g/L.

So can anyone educate me on which culturing medium is the best? Or at least provide a list of top candidates of culturing medium.

Note: I just want to know my bacteria's IAA production ability under normal/standard conditions, so no salinity/heavy metal stress test will be involved.
