I am first time working with Pythium sp. I got few media from research articles such as V8 agar, Corn meal agar, NARF, NARM etc. I want to know which one is better to use for the growth of Pythium ultimum?
Also, you can use PDA medium at a temperature degree of 25 C, but without adding the antibiotic Chloramphenicol, because it prevents the fungus growth.
If you want to isolate the fungus Pythium from the infected plant roots, you can prepare a water culture by putting pieces of roots in these plates and incubating them for 2-3 days. After that, when you see the mycelium, you must transfer it to plates containing sterile PDA medium (without antibiotic) and incubating the plates at 25 C for 3-4 days in incubator.
V-8 Agar medium is also used successfully for isolating Pythium. This medium consists of 8 types of vegetable including carrot, spinach, tomato, celery, parsley, beets, watercress and lettuce. They are mixing together with CaCO3 with known percent and preparing a juice from them.
