Hi,

this is the protocol we used for keratinocyte cultures (normally this was followed by an additional coating with collagen).

poly-L-Lysin coating:

wash coverslips with Ethanol (100%)

wash coverslips H2O

let coverslips dry

coat coverslips for 5 min with120ul poly-L-Lysin solution (MW 70,000-120,000: 0.01%, Sigma)

wash 3 times with H2O

let coverslips dry

Poly(lysine)-coating of coverglass.

Coverslips were incubated overnight in HNO3, washed six times in ddH2O and dried overnight in an oven at 250 °C. Four coverslips were placed in 6-cm dishes and coated with 200 μl of 1 mg/ml poly(lysine) solution (MW 30,000–70,000, Sigma), incubated for 3 h at 37 °C and afterwards washed three times in ddH2O.

After coating, do you also UV-sterilize them before using them for the cells?
