I am applying the Lowry method to characterise the amount of water soluble proteins in permeabilsed cells of Chlorella Vulgaris. Since I used a hard desruptive technology such as High Pressure Homogenisation which allowed to completely release the whole proteic fraction from the microalgae, I would like to characterise them between water soluble and non water soluble fractions. Is there any reference I can look at? Thank you for your kind help.
Do a low-speed centrifugation of the extract (5,000 x g for 10 minutes) to remove large particles, unbroken cells, and cell fragments. Then spin the supernatant in an ultracentrifuge (1 hour at 100,000 x g, for example). The supernatant contains the water-soluble proteins. The pellet contains the insoluble proteins.
