'Modern' is also subject to interpretation. As is the 'highest' - most commercially important? Most significant in raising human happiness? etc.
My own five suggestions, keeping to the 20th century and later;
* Sulfonomide antibiotics
* Penzias & Wilson's discovery of the CMB
* Gödel's incompleteness theorem
* The Haber process
* Directed genetic engineering
