Hello everyone,

I have tried visualizing viral ssRNA (~11kb) on different gels, but no bands were visible with any of them. Before loading the samples onto the gel, I denature them with a denaturing buffer containing 50 % v/v formamide, 20% v/v formaldehyde and 1,5 x MOPS, incubation in 75°C for 5 min and immediate cooling on ice for 5 min.

So far, I have tried:

- 1% agarose gel with 1xTAE (120V 20 min)

- 1% agarose gel with 1x MOPS (120V 20 min)

- 1% formaldehyde agarose gel with 1x MOPS (made from DEPC-water) (60 V, 60 min)

None of them have worked.

Do you have any recommendations or tips?
