Hi, I plan to overexpress a very large bacterial protein (350 kDa). I have some doubt about how e.coli can handle that. Anyone to share some advices? Is it then better to try insect cells or yeast ? Give me your opinion.
E. coli should be able to handle that, it is a bacterial protein initially after all. I would try E. coli first before going to the more complicated systems.
If it doesn't work, Mohammeds answer is very good. We were using both yeast and insect cells to express the dynein heavy chain (~350 kDa) in the Carter lab once upon a time. They each have their pros and cons of course.
