Methanolic extract of cinnamon species is dark and using the disc-diffusion method, the MIC was hard to read. What to do? I'm testing its antifungal activity, and I don't have a Zone Reader. Please help!
Better to make serial dilutions, prepare the discs and after drying use them, this would be fine. But you may also go for spot test or well diffusion method. Both will be working fine.
Diluting your extract to more extent may help. Please try to use extract of low concentration showing better efficacy. I agree with the suggestiins made by Azra Yasmin. You may go for fractionation and islotion of components from the crude extract using column chromatographic technique. It is advisable to work on fractions rather crude extract.
Have you performed Disc diffusion assay or well diffusion assay?
If yes, what exactly is the problem you are facing?
Don't worry if you do not have zone reader. You can simply measure the zone with a ruler (Scale) form different side and then note the average as your reading.
