I was curious about an aspect of cloning and plasmid purification. The typical plasmid purification procedure isolates general plasmid from the cell and is not sequence specific.
So why is it that you don't have a variety of other plasmids in addition to your plasmid of interest? Is it that strains popular in molecular cloning are particularly low in endogenous plasmid so it is less of a factor? Or perhaps is it that high copy number of the cloning plasmid are necessary in a cell to get sufficient expression of the antibiotic resistance gene and thus the levels of endogenous plasmid are insignificant relevant to the high number of cloning plasmid?
