I am staining cells for FACS for cytokines and transcription factors. Is it possible that I postpone the intracellular staining one day without compromising quality of satining?
Yes, it is totally feasible. Just want to add that in your case you have no need for surface marker staining. Just fix the cells, permeabilise them with triton-x or whatever detergent you use. Then after breif washing, block the cells in 0.5% BSA in sheath fluid or PBS. At blocking step, you can leave the cells overnight at 4degree. Come next day and perform your intracellular cytokine staining procedure.
Thanks for the great advice. A colleague of mine experienced problems with T-bet staining when staining fixed cells one day later. Do you stain for T-bet? Have experienced any problems?
