Can an ELISA assay for glucocorticoids (GCs) measurements be used in assaying GCs in tissue lysates?

I would like to measure GCs in tissue samples and I have tissue lysates. Is it feasible to use an ELISA-based assay or is there a better alternative? Thanks in advance!

When staining ex vivo cells for FACS analysis, can I fix the cells and stain them for transcription factors (esp. T-bet) the next day?

I am staining cells for FACS for cytokines and transcription factors. Is it possible that I postpone the intracellular staining one day without compromising quality of satining?
