I've got a recognizing problem with hexaploid wheat hybrids possible translocations in their chromosomes. After FISH, I've obtained in most cases 3 pairs of chromosomes (1B, 6B and 5D) with visible 35S rDNA signals. During karyotype analysis I started to wonder if there is a translocation on 1B (at the beginning I assumed it was possible 1BL/1RS wheat-rye translocation) and 6B or it is just nucleolar organizer region. I'd very grateful if you could help me with this. As you can see, there is not such distance between chromosome fragments on 5D where also 35S rDNA signal was detected.
Identification of these chromosomes was based od repetitive sequences signals and 35S rDNA signal.
High resolution snapping under oil immersion (of 100 x objective lens with eye piece 10 X), multicolor FISH is powerful. We must develop very special skill to identify each chromo arm of the bread wheat. We must have strong logic too for its interpretation which we can gain with special hands on effort and theoretical insight and time. Furthermore, We should work with the hybrid between your BW material line (genotypes) and the CS. We should see meiotic metaphase (M I) too. If it is possible work with multi color FISH with MI chromo spred though it is tough.
There is no transocation between 1B and 6B chromosomes. 1B, 6B and 5D chromosomes contain NOR.
