I want to stain the human TIL with cell marker antibody.When I try to order the antibody..I notice that some of the antibodies named anti-human antibody and some named anti-mouse antibody..It's only for the flow surface staining ..I am super confused and can anyone tell me which one should I order..FITC anti-mouse antibody or anti-mouse antibody?
Sometimes while the mouse and human gene may have the same name, the sequences are sufficiently different that an antibody for one may have a significantly lower affinity for the other. Normally I would suggest checking the homology between the human and the mouse, but in this case, you are staining human tissue and a human antibody is available.
These are the types of secondary antibodies which are raised either against primary human or mouse antibody, respectively. You should order these secondary antibody while considering which primary antibody do you have.
Anti-isotypic genes are present on the Fc regions of antibodies which are species specific. When human sera is administered in rabbit or goat , rabbit or goat antihuman antibodies are produced which anti isotypic and anti idiotypic both.
In research we use mainly antisotypic antibodies which are specific against the Fc fragment such as goat or rabbit antihuman antibodies will bind to Fc region of human antibodies only.
