I'm using the software PAST to compare the diversity of 2 locations. Which T test should I used to compare the locations because Diversity T test gave a significant difference result with a P value lower than 0.05 while Independent 2 sample T test gave a P value above 0.05.

They are the same as long as the groups are unrelated. But 'diversity' t test is not a thing. There are only 3 types:
- An Independent Samples t-test compares the means for two groups.
- A Paired sample t-test compares means from the same group at different times (say, one year apart).
- A One sample t-test tests the mean of a single group against a known mean.
If you are comparing something like Shannon diversity or Simpson diversity, you should use the appropriate "diversity" t-test. The PAST manual should give you some details. The test will be using quantities like Shannon diversity index and species richness, and is distinct from the common independent samples t-test.

Diversity t test is used for comparison of the Shannon and Simpson diversities in two samples. Requires two columns of abundance data with taxa down the rows. But t test is used for comparison of two means.
