I want to ask about the difference between 5-rox and 6-rox dyes, especially about the difference between their spectral properties. When i'm looking in commercional sites where 5 and 6 isomers in stock, in applification notes i see identical spectral properties. It's true?
So, can I use 5-isomer instead of 6-isomer?
If you look at their structures (use ChemSpider) the only difference is the position of a carbonyl group which is remote from the core fluorophore. Although having an electron withdrawing group in a meta position instead of a para position would have a minor impact on the absorption spectra of phthalic acid group (eg. https://pubs.acs.org/doi/pdf/10.1021/ac60140a006 , https://webbook.nist.gov/cgi/cbook.cgi?ID=C100210&Mask=400 and https://www.sciencedirect.com/science/article/pii/S1386142516301615), this is quite minor.
