I recently started working with the VCaP cell line. I used ATCC recommended media to propagate those cells. I am having a hard time propagating them. Can someone recommend the media composition that would be the best to work with this particular cell line?

Culture medium for VCaP cell line is DMEM : Ham's F-12 (1:1) + 2mM Glutamine + 10% FBS.
I would like to mention a few things about this cell line.
VCaP cells have to be treated gently, especially at the time of centrifugation you should use low speed. These cells usually form clumps. Please do not try to break the clumps because they are delicate cells. You will come across a lot of debris in culture. This is normal.
The cells grow very slowly in culture. They take minimum 2-3 weeks to reach confluency in a T-75 flask. Whenever you subculture the cells always make use of conditioned medium. For a 10ml growth medium use 1ml conditioned medium and 9ml culture medium. So preserve the conditioned medium at -20 degree C.
While trypsinizing the cells be careful not to over expose the cells to trypsin because the cells are sensitive to trypsin. Not all cells adhere to the flask, some keep floating in the culture medium. Use of coated flask will help.
