Synovial fluid was already collected from knee joint and firboblast cells were cultured from the collected synovial fluid. What will be the control of synovial fibroblast cells of rheumatoid patient?
Fibrobalsts in RA synovial fluid may come from various zones of tissue and may be in various states of activation or differentiation. Once cultured they may return to a fairly homogeneous state that does not reflect any particular population in vivo.
If you want to compare them with other cell populations the choice depends on what questions you are interested in I think.
Thank you sir for your valuable answer.I am just doing the same thing.Collected synovial fluid from Osteoarthritis patients and thn cultured that cell in vivo condition after that we isolated synovial fibroblast cell and compared this with RA synovial fibroblast.Sir is this procedure is ok?
Sir,i just want to demostrate the mechanism of inflammatory pathways inside the synovial fibroblast cell in Rheumatoid patients.As a control group i am isolating synovial fibroblast cell from primary OA patient.is this ok?
By the time the cells are cultured all the inflammatory signals are likely to have been washed out of the system. The fibroblasts from RA tissue are likely to continue to show some differences in gene expression and differentiation but I am not sure you can conclude anything useful from that.
I would also be careful about the term 'fibroblast cell'. What you have in culture are fibroblasts. There is no guarantee that they derive from the 'fibroblastic synovial intimal cells' or 'type B intimal cells'. They are just as likely to be fibroblasts from deep tissue. There is a lot of confusion in the methodology and terminology in this field.
Synovial fibroblasts cells are playing a key role in pannus formation inside the synovium. In normal condition,in knee joint the number of synovial fibroblast cells are very low,also in diseased condition they are increased inside the synovial fluid.its all about Rheumatoid arthritis pathogenesis.
So sir,could you suggest me how can i framed my research work with this synovial.fibroblast cell? Because without treatment of this particular cells the pannus formation cant be reduced
