What methods are best used in the statistical analysis of ultrathin cell sections for transmission electron microscopy? I need to estimate the number of vesicles 50-100 nm in size in different samples. Unfortunately, other microscopic methods cannot be used here.
I did similar analyzes to estimate the amount of plastoglobules and starch grains in chloroplasts, and measured the thickness pit membrane of vessel elements. Both analyzes using TEM. I obtained images at the same magnification and processed the images in the Image Pro Plus software. Through this software it is possible to measure the quantity, thickness, diameter, area of any sample and structure. The only care you need to take is to standardize the magnification of all images and obtain images with bars to calibrate the software. In addition to this software, there is ImageJ which allows you to do similar meditations. From the values you obtained you evaluate which is the best statistical test to apply to your data, respecting the normality and homecedast of the data. I don't know if I helped you.
